This powerpoint presentation begins by providing the research objective: to better understand the fate ofmanganese - whether from sourcewater or treatment plant impurities -under conditions of biological filtration,and to improve the water industry'sunderstanding of, and ability to control,manganese release or pass-throughduring biological treatment. Biofilm T-RFLP analysis results:T-RFLP analysis has been completed on 32 of the 41samples received, with nine samples failing to amplify byPCR, apparently due to low DNA concentrations and/orPCR inhibitors in the DNA;T-RFLP is continuing on more referencecontrol strains; statistical analysis ofthe T-RFLP profiles; the PCR tests were completed for Pedomicrobiumand Leptothrix; quantitation of PCR results in progress; comparison of T-RFLP and PCR shows that T-RFLPpeak 148 is not a reliable predictor of the presence ofPedomicrobium; comparison of T-RFLP and PCR shows that T-RFLPpeaks 119, 138, and 470 are not reliable predictors ofthe presence of Leptothrix; and,T-RFLP peaks are often characteristic of severalbacteria. The identity of members of the communitycontributing to a peak can only be resolved byspecific PCR or 16S rRNA gene clone libraryanalysis. Includes tables, figures.
